The TRINITY V3 caliper provides backup reading (rarely used) for the automated system and for reading of large single plate assays. The TRINITY V3 caliper records zone diameter measurements instantly at the push of a button through the computer USB port.
Large Single Plate Assay - Overview
The TRINITY V3 caliper can be used with large NUNC (Denmark) bioassay plates. Samples may be laid out based on Latin Square, random, sequential or a custom format. This process takes about two seconds per zone measurement, and the prompt on screen shows the exact zone to measure next. This can be used with any size of petri plate and any layout of sample positions. A standard curve graph is generated and results are automatically calculated with required statistical analysis.
Large Single Plate Assay - Test Results
A standard curve is plotted with the average of standard (known) concentrations. Unknown concentrations are calculated mathematically, based on the standard regression formula derived from the standard curve. The plotted curve shown on-screen to help visualize the on-scale/off-scale values. Correlation coefficient, slope, and intercept are displayed. Test data and results can be saved, printed, and exported.


Large Single Plate Assay - Layout
The layout of test samples is based on having all standard and unknown concentrations on one single plate. This eliminates plate-to-plate variation and facilitates plate setup and reading. Alternatively, two identical plates may be laid out, read and averaged. NUNC (Denmark) large 243mm square plates may be used to enable up to 64 samples to fit on one plate. This provides for up to 6 standards and up to 10 samples, with 4 replicates of each standard and unknown concentration on one plate.
Plates of any size can be used for this type of assay. TRINITY software allows 2-10 standard concentrations and 1-100 unknown samples with up to 8 replicates of each. Sample layout can be Latin square, random, sequential or user defined. User defined layouts can saved for future use.
